Methotrexate is a folic acid antagonist that interferes with DNA synthesis. It acts by irreversibly binding to dihydrofolate reductase, an enzyme involved in the conversion of dihydrofolate to tetrahydrofolate. Tetrahydrofolate is essential for the synthesis of purines and thymidylate, which are building blocks of DNA.
